FOXO4-DRI is a 46-residue D-retro-inverso peptide: it is built from D-amino acids in reversed order, a design intended to resist proteolysis while mimicking the side-chain arrangement of a FOXO4 region. It includes a cell-penetrating sequence and is identified by CAS 2460055-10-9.

What the research establishes
Baar and colleagues designed FOXO4-DRI to interfere with the interaction between FOXO4 and p53 in senescent cells. In culture, the peptide caused p53 to leave the nucleus and triggered apoptosis selectively in senescent cells. In mice, they reported that it counteracted doxorubicin-induced chemotoxicity and improved fitness, fur density and kidney function in fast-ageing XpdTTD/TTD and naturally aged animals.
These findings concern the peptide as prepared and tested in those cell and mouse models. They do not describe the properties or performance of this product, and independent replication of the in vivo results remains limited.
